Older Li-ion cells actually handle more charges, maybe as many as 1,000 full charge-discharge cycles. But they also age independently of the use, dropping between 5% and 20% of total capacity every year, depending on how they're kept (temperatures, charged or discharged, etc).
End of life/failure is really more of a judgment call than a specific event, since, aside from drastic failure (damage or defect), they just get progressively worse. But it happens slow enough you may not notice, unless you're comparing an older cell against a new one. If you start to notice the battery just seems to die sooner than it once did, it's probably about time to think about a replacement.
One thing to watch... Li-ion and Li-poly cells can be damaged by draining them too far. What happens is that a secondary battery more or less forms in the contact area of the cell and starts to kill the contact. So while you really want to fully discharge NiCAD and NiMh cells for the best performance, you shouldn't let a Lithium cell die completely.
